Abstract:
Introduction: Cerium oxide nanoparticles or Nanoceria as radio-protectors can play an important role in reducing the complications of ionizing radiation. Reducing the side effects of 6MV photon beams that are commonly used in radiotherapy can be remarkable. Methods and Materials: Nanoceria suspensions were sterilized with 70% ethyl alcohol. The suspension was shaken by vortex for 3 minutes to optimize the distribution of nanoparticles in aqueous medium and sonication performed by ultrasound sonicator for 2 hours. MRC-5 cells were cultured in DMEM / F12 medium and incubated in high humidity at 37°C. In order to determine the non-toxic concentration, the cells were treated with serial concentrations of 5, 10, 30, 50, 70, 90, 110, 150, 200, 250 and 300 μM of nanoceria. Quantitative radio-protection effect of nanoscale was performed in non-toxic concentrations with radiation doses of 20, 40, 60, 80 and 100 cGy of Megavoltage X-Ray radiation. Results: 70 micromular and lower concentrations of nanoceria were not toxic for MRC- 5 cells. The mean cell viability in this concentration of nanoceria was 89.4 ± 2.6%. MRC-5 cells in presence of 70 μM nanoceria with radiation doses of 40, 80 and 100 centimeters had significant radiation protection compared to the control group (P-Value˃0.005). Discussion and Conclusion: The use of cerium oxide nanoparticles can increase the accuracy of treatment and decrease the secondary effects of radiotherapy.
